Pharmacological action

Pharmacodynamics

The antibacterial action mechanism of penicillin is mainly to inhibit the synthesis of bacterial cell wall sticky peptide. Penicillin has mainly role in killing a variety of gram-positive bacteria and minority gram-negative bacteria. The main sensitive bacteria of  penicillin are staphylococcus, streptococcus, bacillus erysipelatos-suis, corynebacterium, clostridium tetani, anthrax bacillus, spirochaeta,etc. Penicillin is not sensitive to mycobacteria, mycoplasma, chlamydia, rickettsia, Nocardia bacteria, fungi and viruses. Dihydrostreptomycin sulphate belongs to aminoglycoside antibiotics, its mechanism to inhibit bacterial protein synthesis, increase cell membrane permeability to induce bacterial death, it has antibacterial activity on mycobacterium tuberculosis and a variety of gram-negative bacilli ( Such as E. coli, salmonella, brucellosis, pasteurella, shigella, bacillus mallei,etc), it has poor antibacterial activity on most gram-positive bacilli (such as Staphylococcus aureus).

Pharmacokinetics

This product is well absorbed after intramuscular injection, it will reach peak plasma concentration about 0.5-2 hours, eliminate slowly, high bioavailability. Penicillin can maintain a lower effective plasma concentration more than 12 hours. Streptomycin sulfate, peak concentration can reach 51μg / ml, can kill the sensitive bacteria effectively, can pass through the placental barrier,  is mainly excreted by the kidneys.

Drug Interactions

1. Quick effective antibacterial agents (such as macrolides, tetracyclines and amide alcohols) have interference effect on the bactericidal activity of the product, it should not be used in combination.
2. The activity of this product can be damaged by heavy metal ions (especially copper, zinc, mercury), alcohols, acids, iodine, oxidizing agents, reducing agents, hydroxyl compounds, acid, glucose injection or tetracycline hydrochloride injection, this belongs to compatibility taboo.
3. This product should not be mixed with some drug solution (such as chlorpromazine hydrochloride and hydrochloric acid Lin can mycin, tartaric acid to norepinephrine (NE), oxytetracycline, tetracycline hydrochloride, group B vitamin and vitamin C), otherwise can produce turbidity and floc or precipitation.
4. This class of drugs will enhance antibacterial activity in an alkaline environment, it will strengthen antibacterial effectiveness, when it to be used in combination with alkaline drug (such as sodium bicarbonate, aminophylline, etc.), but the toxicity is correspondingly enhanced.
5. Ca2 +, Mg2 +, Na +, NH4 + and K + and other cations inhibit the antibacterial activity of this class of drugs.
6. It can enhance the ototoxicity of this class of drugs to combine with cephalosporins, dextran, potent diuretics (such as furosemide, etc.), erythromycin.
7. It can enhance the neuromuscular blockade of this class of drugs to combine with skeletal muscle relaxants (eg succinylcholine chloride, etc.) or a drug that has such effects.

Indication:

It can be used to treat a variety of infections or mixed infection caused by sensitive bacteria, such as various kinds of diarrhea, mammitis, hysteritis, septicemia, cystitis, Cattle haemorrhagic septicemia, calf pneumonia, swine plague, leptospirosis, fowl cholera, swine brucellosis,horse pyohemia sepsis (suppurative nephritis and arthritis), foal pneumonia, tuberculosis,etc.

Usage and Dosage:

Intramuscular injection

Shake well before use

Livestock: 1ml per 25kg bodyweight, daily, during 2～3days

Adverse reactions:

1). Most livestock may have an allergic reaction, but the incidence is lower. Local reactions showed edema and pain at the injection site, systemic reaction are urticaria and rash, severe cases can cause shock or death.

2). Ototoxicity is relatively strong, vestibular dysfunction often happens, it will increase with the accumulation of continuous administration of drugs and show a dose-dependent manner.

3). This product overdose leads to neuromuscular blocking effects.

4). Long-term use can cause kidney damage.

Precautions

1). This product is forbidden use for patient livestock, which has penicillin, procaine and aminoglycoside drug allergy.

2). Patient livestock is dehydration (which can cause increased plasma concentration) or impaired renal function when used with caution.

Withdrawal Period：Pigs, cattle, sheep～28days
